Theoretical part:
- Introduction to animal models – spontaneous mutants and transgenics
- History of transgenesis
- Transgenesis methodology
- DNA constructions
- Genotyping the founders
- Transgenesis applications
Practical part:
- Visit to CEDEME
- Mice lines used to produce transgenics
- Breeding and preparing the animals to generate a transgenic line
- Vasectomy of adult males
- Preparing the foster mothers
- Superovulation of donor mother
- Preparing the vasectomized males
- Checking pregnancy by vaginal plug
- Collecting embryos in different phases of development
- Preparing the DNA for microinjection
- Pronuclear microinjection
- Preparing stem cells for injection
- Injecting stem cells into blastocysts
- Embryo transfer
